His father, John Jefferson, played NCAA Division II basketball, while his brothers, Jordan and Rickey, also played college football. Justin attended Destrehan High School, where he excelled in both football and track. His impressive performance in high school led him to commit to Louisiana State University (LSU).

He continued to impress with 88 receptions and seven touchdowns, earning a spot on the PFWA All-Rookie Team and a Pro Bowl selection.
